Anti-tumor efficacy and biomarker evaluation of agonistic anti-OX40 antibodies in preclinical models

چکیده
Background OX40, a TNFRSF member, is a co-stimulatory molecule expressed on antigen experienced effector T (Teff) and regulatory T (Treg) cells, including infiltrating cells in mouse and human tumors. Activation of OX40 by agonistic antibodies is hypothesized to promote anti-tumor immunity by enhancing Teff activation and inhibiting Treg mediated suppression. We conducted in vitro and in vivo preclinical studies to characterize co-stimulatory activity, evaluate anti-tumor efficacy and identify predictive and pharmacodynamic (PD) biomarkers of agonistic anti-OX40 antibodies.

Fc Engineering Approaches to Enhance the Agonism and Effector Functions of an Anti-OX40 Antibody*
Agonistic antibodies directed against immunostimulatory receptors belonging to the tumor necrosis factor receptor (TNFR) superfamily are emerging as promising cancer immunotherapies. Several Fc engineering approaches discovered recently can augment the anti-tumor activities of TNFR antibodies by enhancing their agonistic activities and/or effector functions. In this study, we compared these app...
متن کاملApoptotic and antitumor activity of death receptor antibodies require inhibitory Fcγ receptor engagement.
By virtue of their ability to induce apoptosis and regulate growth, differentiation, and cytokine responses, the tumor necrosis factor receptor (TNFR) superfamily members have emerged as attractive targets for anticancer therapeutics. Agonistic antibodies to apoptosis-inducing TNFRs, such as death receptor 5 (DR5), although displaying impressive activities against a variety of tumors in preclin...
متن کاملTherapeutic antitumor efficacy of anti-CD137 agonistic monoclonal antibody in mouse models of myeloma.
PURPOSE Eradication of post-treatment residual myeloma cells is needed to prevent relapses, and immunostimulatory monoclonal antibodies (mAb) such as anti-CD137, CTLA-4, CD40, etc., which enhance the immune response against malignancies, represent a means of achieving this purpose. This study explores anti-CD137 mAbs for multiple myeloma treatment in preclinical models of the disease because th...

The blockade of immune suppression against antitumor responses is a particularly attractive strategy when combined with agents that promote tumor-specific CTLs. In this study, we have attempted to further improve the CTL induction and potent antitumor efficacy of a combination mAb-based therapy (termed "trimAb therapy") that comprises tumor cell death-inducing anti-death receptor 5 mAb and immu...
متن کاملAnaphylaxis caused by repetitive doses of a GITR agonist monoclonal antibody in mice.
Immunotherapy for cancer using antibodies to enhance T-cell function has been successful in recent clinical trials. Many molecules that improve activation and effector function of T cells have been investigated as potential new targets for immunomodulatory antibodies, including the tumor necrosis factor receptor superfamily members GITR and OX40. Antibodies engaging GITR or OX40 result in signi...
